I think the set of instructions in the tweet is simply the map he's referring to:

We've been following it so far, and he does refer to that tweet as being "our map" so I'm not sure the Witness puzzle will be useful here. Ya never know though, any ideas on how it's implemented?

For now Ed has basically spelled out our instructions for us. We need to use the pieces to decode the numbers somehow.
